Amelia Peabody from Elizabeth Peters' books. Though it's hard to keep in mind the difference between favourite series and favourite 'lady detective'. For instance, I like Donna Andrews' Meg Langslow series as a whole, but Meg's only a part of that, where Amelia Peabody is a much more important part of why I like that series of books.
